Yeovil are yet to win under manager Terry Skiverton and Jones, but are unbeaten at home. The Glovers enter the match at Huish Park in the Coca-Cola League One relegation zone, two points from safety.
The Robins' dramatic 4-3 victory against Northampton Town on Tuesday has seen Danny Wilson's team move from 20th to 15th in the table, five points ahead of Yeovil.
Jones said that the squad had benefited from a week of training uninterrupted by games.
He said: "It is as big a game as you can get for us. It is a game we are looking forward to and one that is against a side that if we a get a good result against it will be doubly as good; not only do you pick up points yourself, but you stop others around you picking up points.
The left back who has made more than 130 appearances for Yeovil said himself and Skiverton had done their homework on Swindon and were well aware of their capabilities.
As well as being in the crowd for Swindon's 3-0 demolition of Southend United last week, Jones travelled to the Sixfields Stadium for the Robins' Tuesday night fixture. Skiverton's number two highlighted 24-goal striker Simon Cox as one of Swindon's main threats ahead of Saturday's match.
"They are in form and I went to watch them last week against Southend United when they had a good result," said Jones. "They had another good result at home on Saturday against Hereford United. We know enough about them and we know they have some good players. Cox up front is a quality player and is in a good vein of form at the minute. It is a challenge that we are relishing and looking forward to."
Jones added that he and Skiverton will have an almost-full squad to choose from for the visit of Swindon.
